Specificity: This item detects Notch 2, an important signalling protein which is critical for embryonic development. Notch 2 is a type 1 transmembrane protein that is activated by Delta1, Jagged1 and Jagged2. Activation results in cleavage and translocation of the Notch intracellular domain (NICD) to the nucleus, where it functions as a transcriptional regulator.
Notch 2 activation results in differentiation and proliferation in many cells, in particular in myeloid cell differentiation, functioning in the maturation of CD8 thymocytes and the development of Marginal Zone B cells.
Recently Notch 2 mutations have been shown to exist in a minority of patients suffering from Alagille syndrome (AGS), a dominant, multi-system disorder. Notch 2 mutations can also be oncogenic, for example in B-cell chronic lymphocytic leukaemia (B-CLL).
